Does anyone know if there is a way to email a response directly to a comment left on a blog that uses the new Blogger? I'm thinking the answer is "no", but thought I would ask in case there's something I'm missing.

I don't think there is because there is nowhere to add an email address if you are not posting a comment via your blogger account.
If the comment was made by someone with a blogger account I guess you can get their email from their profile if they give it.
On Typepad there is a field for email address when you leave a comment so your name either links to that or your blog if you give it.
